Hydrochloride procaine is a local anesthetic that temporarily blocks nerve fiber conduction to produce an anesthetic effect. It boasts potent efficacy, low toxicity, and no addictive properties. However, its ability to penetrate the skin and mucous membranes is weak, making it unsuitable for surface anesthesia. Clinically, it is primarily utilized for infiltration, spinal, and conduction anesthesia.The aqueous solution of this agent maintains relative stability within a pH range of 3.0 to 5.0. Thus, when preparing hydrochloride procaine injection solutions (as noted in Chemicalbook), acid is generally added to adjust the pH to 3.3–5.5, with strict control over sterilization temperature and duration. Sterilization via 100°C circulating steam for 30 minutes is recommended, and the product should be stored in a sealed, light-protected container.The aromatic amino group in its chemical structure reacts with sodium nitrite in dilute hydrochloric acid to form a diazonium salt. This salt then undergoes a coupling reaction with alkaline β-naphthol, resulting in the formation of a scarlet azo compound.
